Deck calibration? No. You only need to deck calibration in certain cases. See: deck calibration

Pipette calibration? Yes. You must calibrate your pipette every time you run a protocol. It is quick and easy.

Labware calibration? It depends. 

If you have run that protocol before (with the exact labware set up) and saved the calibration to the robot, then 'No.' 

The calibration for that labware was saved. If you are running a new protocol, then you must calibrate the labware. For labware calibration, calibration persists across protocols; so, if you load a new protocol, it will automatically use the last calibration you saved.
